520 _ _ |a Inositols (cyclohexanehexols) comprise nine isomeric cyclic sugar alcohols, several of which occur in all domains of life with various functions. The most abundant isomer is myo-inositol (MI). Its rare isomers, scyllo- (SI) and D-chiro-inositol (DCI) are promising drug candidates fortreating Alzheimer’s disease, diabetes type 2 and polycystic ovary syndrome. Therefore, cost efficient processes for the production of these compounds are desirable. Many bacteria can utilize inositols as carbon and energy source via a specific pathway involving inositoldehydrogenases (IDHs) as the first step of catabolism, followed by the actions of inosose isomerases. The microbial cell factory Corynebacterium glutamicum can grow on MI as sole carbon source and possesses many uncharacterized genes that are annotated to contribute toinositol degradation. It also has the innate ability to synthesize MI from glucose-6-phosphate. This thesis aimed to elucidate the function of the undescribed genes for inositol metabolism and exploit the potential of C. glutamicum to be engineered as a suitable host for the biotechnological production of SI and DCI.
