AB - In this work a system of culture techniques was developed for the ex vivo expansion of hematopoietic cells, which is of major importance in cancer therapy. For stroma dependent cocultivation, the fixed bed reactor and the 1 mL miniaturized loop reactor allow the cultivation for clinical and screening purposes, respectively. Here the cells are immobilized on macroporous carriers . For a suspension culture a reactor was developed that has a working volume between 40 and 550 mL. An integrated filtration allows a continuous cultivation up to densities of 1 . 107C/mL. Finally, a small scale spinner vessel of 10 mL suspension offers the possibility of a screening in suspended culture. Additionally, some relevant parameters for the cultivation of hematopoietic progenitor cells were investigated. It was shown, that the materials the cultivation systems are made of have an influence on the cultivation, but in technical systems it is to small to be relevant . An optimal osmolality of the culture was found at 0,31-0,32 Osmol/kg . Besides the progenitor cells, T-lymphocytes were cultivated as an example of a more differentiated cell type . After the successful cultivation in the new suspension reactor system, a standard 20L stirred vessel was used to obtain a clinically relevant final cell number of 2,9 - 101° cells. The new suspension systems was used also in a clinical study phase I/II for cancer therapy of children.
