Label-free aptamer – based detection of Amyloid Beta proteins on SiO2 covered silicon structures

Abstract: Neurodegenerative processes in brain became important challenge with increased age of population. Usually relatively bulky optical methods are used to detect Amyloid β peptides, which are linked to the pathogenesis of Alzheimer disease. To prolong effective and healthy memory and thinking capability as well as to stop the factors resulting in Alzheimer’s disease new high-sensitive early-stage methods for detection of peptides involved in Alzhemer’s disease have to be developed. In this contribution we report on successful demonstration of new label-free approach for an aptamer-based detection of amyloid beta proteins. A high-selective nucleic acid aptamer was synthesized and immobilized onto SiO2 surface for detection of the target Amyloid beta-40 peptide. Functionalized structures on thin film dielectric layer has been developed to detect ultra-low concentrations of target protein. The detection technique utilizes high selection aptamer immobilization on the SiO2 dielectric layer of system configured with a liquid-gate. The introduction of amyloid beta protein to the sensor surface of different concentration in a wide range from 0.1pM to 1uM results in change of surface potential. These experimental data provide a sensitive detection of ultra-low concentrations of an amyloid beta protein and opens perspective for the development of real-time sensors for studies and understanding of different stages of Alzheimer’s disease utilizing nanostructures fabricated on the basis of cost-efficient CMOS compatible technology.Keywords: Alzheimer, Therapeutics, Flat-band Voltage, Biosensor, Aptamer, nucleic acid, Amyloid Beta, Ab-40, Debye length, Immobilization, Silicon Oxide, Thin Films
