Abstract: Blood flow in the microvascular system presents a complex interplay between the non- Newtonian properties of blood and the intricate, tortuous architecture of microvessels. While 2D microfluidic systems have enabled detailed studies of red blood cell dynamics, recentfindings by A. Gholivand et al. [1] revealed critical differences in flow behavior between inplane and out-of-plane branches of microfluidic bifurcations. This emphasizes the need for 3D geometries to accurately replicate physiological blood flow.Building on this, we investigate red blood cell flow within non-planar 3D microfluidic networks, uniquely reconstructed from real brain vasculature images and fabricated via selective laser-induced etching. By testing various hematocrit levels and aggregation factors, we aim to elucidate the influence of blood composition on flow dynamics in the brain's complex vasculature. These insights will deepen our understanding of cerebral microcirculation, with potential implications for conditions involving vascular abnormalities.
