Abstract: Umbilical cord blood is a promising source for hematopoietic stem cells (HSC) that can be used as an alternative to bone marrow. As the number of HSC in umbilical cord blood is limited, transplantation in adults is currently still a critical issue. Thus, a possibility to expand HSC from umbilical cord blood via ex vivo-cultivation is needed. For this, co-culture of HSC with feeder cells is a good possibility, because this mimicks the physiological environment of HSC. Up to now, most in vitro-studies used murine cell lines as feeder cells which poses problems for clinical application because of their xenogenic origin. To overcome these, this work aims to provide and compare several human primary cell types as alternative feeder cells. Namely, mesenchymal stem cells (MSC) from bone marrow and umbilical cord blood, human umbilical cord vein endothelial cells (HUVEC) and cells from the umbilical cord matrix tissue (Wharton’s jelly-cells = WJC) were included. All these potential feeder cells were tested mainly with respect to their possible clinical application. The parameters used were e.g. availability of the potential feeder cells, their distinction from HSC and other hematopoietic cells derived from these, the feeder cells’ survival in a clinically applicable culture media optimised for use with HSC, the activation of allogeneic T-cells and the support for HSC-expansion by the potential feeder cells. It could be shown that all potential feeder cell types might be used in a clinical setup. Hereby, their good support for expansion of hematopoietic progenitors is of paramount importance. Especially MSC and WJC supported the expansion of CAFC to a high extent. Additionally, WJC demonstrated potential for other cell therapeutic applications because of their easy isolation and high proliferation rates.
