Abstract: Introduction: [18F]T807 (7-(6-[18F]fluoropyridin-3-yl)-5H-pyrido[4,3-b]indole) is a positron emission tomography (PET) tracer used for imaging pathological tau aggregates, which have been implicated in neurological disorders such as Alzheimer's disease and traumatic brain injury [2]. Although the preparation of [18F]T807 on standard synthesis modules has been successfully established [3-5], cassette-based systems could offer greater adaptability for implementing the radiosynthesis across diverse synthesis sites. In the present work, we describe a robust and GMP-compliant cassette-based procedure for the automated radiosynthesis of [18F]T807 on a Trasis AllinOne (AIO) synthesizer. Methods: [18F]T807 ([18F]3) was prepared by an optimized two-step protocol, involving nucleophilic radiofluorination of the t-Boc-protected nitro precursor 1 and subsequent acidic deprotection of the radiolabeled intermediate [18F]2 (Figure 1). The automated process began with the elution of [18F]fluoride from a QMA cartridge with a solution of Kryptofix®2.2.2 and K2CO3, followed by evaporation of the solvent at different temperatures. A solution of precursor 1 (1.5 mg, 3.8 μmol) in DMSO (0.8 mL) was then transferred to the reactor containing the dried [18F]fluoride and the 18F-for-NO2 exchange reaction was allowed to proceed for 5 min at 110 °C. After cooling to 60 °C, the t-Boc protecting group was removed by addition of 3 M HCl (500 μL) and heating to 100 °C for 5 min. The reaction mixture was then neutralized with NaOH solution, loaded onto an HLB-cartridge, and eluted directly into an HPLC system for purification of [18F]3. Results: The average activity yield of [18F]T807 produced using the AIO system was 19 ± 5.4 % (n = 178), with an overall synthesis time of about 80 min (including HPLC purification). In a single production batch starting from 35-50 GBq, the process yielded 6-9 GBq of [18F]3 with a radiochemical purity of > 99 %. Quality control tests confirmed full compliance with the acceptance criteria defined by the European Pharmacopoeia specifications for 18F-labeled radiotracers. Conclusions: This long-term study demonstrates that the cassette-based Trasis AIO system provides a reliable and GMP-compliant platform for the efficient and reproducible production of [18F]T807.
