Abstract: Introduction: Radiolabeled (het)aryl fluorosulfates [1] and, in some cases, sulfamoyl fluorides [2] can be efficiently prepared via SuFEx-based 18F/19F isotopic exchange. However, application of this approach for the preparation of aliphatic sulfamoyl [18F]fluorides ([18F]ASAFs) has resulted in unacceptably low molar activities (Am). Therefore, the aim of this work was to develop a convenient protocol for the production of no carrier added (n.c.a.) [18F]ASAFs. Methods: Sulfamoyl imidazolium triflates were prepared in three steps by fluorosulfurylation of the corresponding secondary amines with des-Me-SuFEx-IT [3], followed by SuFEx reaction with imidazole in the presence of Ca[N(Tf)2]2 and quaternization of the intermediates with MeOTf. For radiolabeling according to the “minimalist green” protocol [4] [18F]fluoride was trapped on a QMA cartridge, the cartridge was washed with EtOH and the [18F]fluoride was eluted with a solution of the corresponding imidazolium precursor (1 μmol) in EtOH (1 mL). The reaction mixture was then heated at 80 °C for 15 min to obtain the corresponding [18F]ASAFs. The developed protocol was applied for the automated radiosynthesis of an 18F-labeled derivative of the antidepressant amoxapine on a GE Tracerlab FX N Pro module. The hydrolytic stability of [18F]ASAFs was assessed using 4-phenylpiperidinesulfamoyl [18F]fluoride as a model compound. Results: Ten imidazolium salt precursors were prepared in 9–83% yields (Ø = 52%) over three steps. Subsequent radiofluorination according to the “minimalist green” protocol resulted in [18F]fluoride elution efficiencies of 79±5% and afforded the corresponding [18F]ASAFs with radiochemical conversions of 90–99%. The 18F-labeled amoxapine derivative was obtained in an activity yield of 26% and an Am of 69 GBq/μmol (771 MBq tracer) within 1 h. 4-Phenylpiperidinesulfamoyl [18F]fluoride demonstrated complete stability across the entire range of evaluated pH values (2–12) and in phosphate-buffered saline (PBS, pH 7.4) for a minimum of 2 hours. Conclusions: The "minimalist green" approach enables efficient preparation of n.c.a. [18F]ASAFs from imidazolium salt precursors with benign EtOH as reaction solvent and without the need for additives or evaporation steps. This streamlined protocol is particularly suitable for the rapid automation of [18F]ASAFs production, since it requires minimal precursor amounts and offers exceptional simplicity.
